Purpose: Demonstrate a workflow for metabolite detection and structure elucidation by HRAM fragmentation-directed secondary analysis using multiple fragmentation techniques.
Methods: Survey analysis of metabolism samples was performed by HRAM full scan and HCD MS2 analysis on a Thermo Scientific™ Q Exactive™ hybrid quadrupole- Orbitrap mass spectrometry. These analysis were processed using fragment ion searching (FISh) to detect structurally related components in the samples for acquistion of MSn data on a Thermo Scientific™ Velos Pro™, dual-pressure linear ion trap mass spectrometer utilizing a combination of HCD and CID fragmentation techniques.
