Chen J, Pannell L, Schambeau L, Rocker J, Koncar G, Kiyonami R, Waddell K. ASMS 2014 Poster Human pancreatic juice digest was analyzed on an integrated protein identification and targeted quantification platform using nano HPLC coupled with a Thermo Scientific™ Q Exactive™ hybrid quadrupole-Orbitrap™ MS. With only 5 ng of sample, ~1300 proteins were identified. Twenty identified peptides were selected for quantitation using three MS acquisition methods, including MSX-tSIM, MS-PRM, and MSX-PRM. The sensitivity and reproducibility of these research methods are evaluated. Results here establish a baseline to guide MS-based quantification method development for research. The approach has been used for the differential analysis of pancreatic juice samples between healthy and pancreatic cancer subjects in clinical research.
