Purpose: To investigate performance for quantitative assay of 1_,25-Dihydroxyvitamin D3 (1,25(OH)2D3) for research applications using Velos dual linear ion trap mass spectrometer (MS), ExactiveTM ultra-high resolution MS and TSQ VantageTM triple quadrupole MS. Triple quadrupole mass spectrometers are usually the instruments of choice for quantitative determination of 1,25(OH)2D3. The current methods require extensive sample preparation with pre-concentration and derivatization leading to inconsistent results.
Results: In analysis of non-derivatized 1,25(OH)2D3 both, LTQ Velos MS and Exactive MS deliver LOQ of 50 pg/mL. At this concentration signal-to-noise ratio observed on Exactive is about 10 times higher than on Velos. Linearity range for both MS platforms is 50-10000 pg/mL. The Vantage triple quadrupole allows to achieve LOQ of 100 pg/mL with linearity range 100-10000pg/mL for non-derivatized analyte.
