Purpose: Demonstrate effective automatic calibration and optimization of a variety of important parameters related to the Thermo Scientific™ Orbitrap™ mass analyzer Methods: An evolutionary algorithm is employed for optimizing several important ion transmission parameters simultaneously. Simple one-device optimization curves are used when one parameter is known to have a dramatic influence on the results of the optimization. We also show how a novel scoring metric can help alleviate the complexity of the optimization curve for isotope ratios in particular. Results: We show that using a multi-dimensional optimization algorithm in conjunction with appropriately chosen scoring metrics can effectively and reliably optimize transmission of ions into an Orbitrap mass analyzer, resulting in good isotope ratios without compromising signal-to-noise.
