Endogenous metabolite profiling is an extreme example of multi-component quantitation analysis. Tripe Quads are considered the gold standard for Quantitation using selected reaction monitoring. However, as the number of analytes increase SRM method development becomes more labor intensive. Using shorter SRM dwell times to accommodate more components compromises sensitivity and precision. In contract, ultra-high resolution LC-MS provides a highly sensitive and relatively unbiased quantitative assessment of metabolomics samples, without the need for MS-MS; it also obviates the need for multiple platforms for profiling and validation.
